The AVRISP JTAG ICE 2-in1 uses virtual comport to communicate with AVRStudio, so the first step is to loaded the USB driver for it.
1. Confirm the USB driver is loaded correctly.
**Here is COM4 assigned to programmer. But it can be COM2-COM8
2.From the AVR Studio, select AVRISP mode connection
OR
3.From the AVR Studio, select JTAG ICE mode connection
