I am having trouble reading a 27128 chip from a Nissan ECU. I have the Enhanced Willem programmer. Everytime I read the chip the data is different. Looking at certain parts of the data, some of it is correct or mostly correct and other times completely wrong. When I try to do the verify it always gives me an error quickly. I desoldered the chip out of the ECU as best I could, maybe this chip got damaged? Or am I doing something wrong? Thanks.

I will mainly be flashing an AT29C256. I only need to read the 27128 chip once, is it at all possible?

Yes, it is possible. I think you need to route the power source by a wire.

Does anyone know what wires I need to cross to read this chip?

The 27128 is a 28 pin DIP chip. The Vcc pin is on pin 28. So, cross the pin 28 and ZIF 32. see if this works.
